Triclofos is a sedative-hypnotic drug primarily used for the short-term treatment of insomnia and as a premedication for sedation in children. It is a prodrug that is metabolized in the liver to trichloroethanol—the same active metabolite as chloral hydrate—producing its sedative effects. Triclofos acts mainly by enhancing gamma-aminobutyric acid (GABA) activity at GABA receptors in the central nervous system (CNS), leading to neuronal inhibition and sedation. Its onset of action and palatability are considered superior to chloral hydrate with less gastric irritation. The drug has been used as an alternative or adjunctive agent for nausea, vertigo, labyrinthine disorders, anxiety or tension before medical procedures. Triclofos has largely fallen out of use in many countries due to safety concerns and availability of safer alternatives; it is no longer available in the United States[1][3][7].
