A therapeutic cancer vaccine platform consisting of a recombinant viral vector (either vaccinia or fowlpox) engineered to express the tumor-associated antigen carcinoembryonic antigen (CEA) with an enhancer agonist epitope (designated 6D), along with a TRIad of COstimulatory Molecules (B7.1, ICAM‑1, and LFA‑3; collectively called TRICOM). The addition of these costimulatory molecules is designed to enhance antigen-specific T-cell responses and antitumor activity by improving immune recognition and activation against CEA-expressing tumors. The platform has been evaluated in phase I clinical trials for patients with advanced cancers expressing CEA, including colorectal cancer and other carcinomas. It is developed primarily by the National Cancer Institute[1][2][4][7].
