The TRICOM vaccine is a therapeutic cancer vaccine platform based on recombinant poxvirus vectors engineered to express a triad of human T-cell costimulatory molecules—B7-1 (CD80), ICAM-1 (CD54), and LFA-3 (CD58)—alongside tumor-associated antigens. The inclusion of these three costimulatory molecules synergistically enhances antigen-specific T-cell responses against tumor antigens. Several specific vaccines have been developed using the TRICOM platform: - PROSTVAC (PSA-TRICOM): targets prostate-specific antigen for prostate cancer. - PANVAC (CEA-MUC1-TRICOM): targets carcinoembryonic antigen and MUC1 for various carcinomas. - MVA-BN-brachyury-TRICOM: targets brachyury in advanced cancers. These vaccines are designed to stimulate robust anti-tumor immune responses by improving the activation and proliferation of cytotoxic T-cells against cancer cells expressing the target antigens. The platform has shown safety and immunogenicity in multiple phase I/II clinical trials across several solid tumors[2][3][5][9].
