Overview

TriCON (Triple Convergent Oncology Nanotherapy) is an experimental carbon-based, triple-modal nanotherapeutic platform designed for the treatment of pancreatic ductal adenocarcinoma (PDAC). The platform is engineered to address the highly malignant and immunosuppressive tumor microenvironment of pancreatic cancer through three synergistic mechanisms: CRISPR/Cas9-mediated gene editing of the poliovirus receptor (PVR/CD155), nano-encapsulated doxorubicin chemotherapy, and checkpoint blockade immunotherapy. TriCON is characterized by spatiotemporal convergence and stimuli-responsive controllability, allowing for precise release of its therapeutic payloads within the tumor. By knocking down PVR, the platform reverses tumor-induced immunosuppression and activates natural killer (NK) cells, while doxorubicin induces immunogenic cell death (ICD), collectively promoting robust antitumor activity and tumor regression in preclinical models.
