Overview

Trifarotene is a fourth-generation topical retinoid indicated for the treatment of acne vulgaris in adults and children aged 9 years and older. It is the first retinoid to selectively target the retinoic acid receptor gamma (RAR-γ), which is the predominant RAR isoform in human skin. By acting as a potent and selective agonist of RAR-γ, trifarotene modulates gene expression involved in cell differentiation, inflammation, skin hydration, cell adhesion, and proteolysis. This results in comedolytic (unclogging pores), anti-inflammatory, and depigmenting effects that improve acne lesions and overall skin texture. Trifarotene was first approved for use in the United States in October 2019 under the brand name Aklief. It has also received orphan drug designation for congenital ichthyosis[1][3][5][6][7][8].
