Overview

Trifluridine is a fluorinated pyrimidine nucleoside analog and nucleoside metabolic inhibitor. It acts as an antiviral agent, primarily used in ophthalmic solutions for the treatment of primary keratoconjunctivitis and recurrent epithelial keratitis caused by herpes simplex virus types 1 and 2. Mechanistically, trifluridine inhibits viral DNA synthesis by incorporating into viral DNA in place of thymidine, thereby disrupting DNA replication. In addition to its antiviral use, trifluridine is also a component of the oral combination chemotherapy drug Lonsurf (trifluridine/tipiracil), where it serves as an antimetabolite that interferes with cancer cell proliferation by incorporation into tumor cell DNA[1][3][4][9].
