Overview

Triglu‑5‑formyl‑tetrahydrofolate is a polyglutamated derivative of folinic acid (also known as leucovorin or 5‑formyltetrahydrofolate), in which three glutamate residues are attached to the folate core. It is a member of the folic acid and derivatives class and acts as a coenzyme in one-carbon transfer reactions essential for nucleotide biosynthesis and amino acid metabolism. Polyglutamation increases its cellular retention and binding affinity to enzymes involved in the folate cycle such as serine hydroxymethyltransferase. The triglutamate form is physiologically relevant for intracellular activity but is not used directly as a pharmaceutical agent; rather it represents an active metabolite or storage form within cells[1][9][7]. Its mechanism involves serving as a substrate or cofactor for enzymes that mediate one-carbon transfers critical for DNA synthesis and repair.
