Overview

TriGlytza is an oral combination therapy developed by Myopharm Limited for the treatment of type 2 diabetes mellitus (T2DM). It is a fixed-dose combination consisting of three small molecule drugs: celecoxib (a COX-2 inhibitor), valsartan (an angiotensin receptor blocker), and metformin (a biguanide). The therapy is specifically designed to target multiple distinct and overlapping pro-inflammatory signaling pathways along the RAS-IL1b-Cox2-PGE2-EP3 axes. These pathways are implicated in the progressive deterioration of pancreatic beta cell function and the development of insulin resistance, which are the core defects in T2DM. By addressing these underlying inflammatory drivers, TriGlytza aims to preserve beta cell mass and improve glycemic control more effectively than metformin monotherapy. It is currently being evaluated in Phase II clinical trials in Australia and the United States.
