Overview

A trilocked photodynamic senolytic inducer (PDSI) is an experimental, liver-targeted, type-I photodynamic agent designed to selectively eliminate senescent cells for the treatment of liver fibrosis[1][5]. Upon activation by near-infrared (NIR) light, the trilocked PDSI produces superoxide anions (O2•–) within the lysosomes of senescent cells, triggering immunogenic necroptosis—a form of programmed cell death accompanied by immune activation. This process not only clears senescent cells but also enhances dendritic cell maturation and recruits cytotoxic T cells, potentiating further immune-mediated removal of senescent cells. The agent is further distinguished by its NIR fluorescence and photoacoustic imaging capabilities, allowing high-contrast, noninvasive visualization of senescent cells in vivo. Developed as a strategy for hepatic fibrosis resolution, the trilocked PDSI is preclinical and not yet in human clinical trials[1][5].
