Trilomer-PTK7 conjugate is a peptide-drug conjugate (PDC) developed by Trilo Therapeutics for the treatment of PTK7-expressing cancers. It utilizes the proprietary Trilomer® platform, which consists of mirror-image D-amino acid cyclic peptides. These peptides are designed as bivalent or biparatopic binders that are highly resistant to protease degradation and exhibit low immunogenicity. The conjugate functions by binding to Protein Tyrosine Kinase 7 (PTK7) on the surface of cancer cells and delivering a conjugated cytotoxic or radionuclide payload to induce cell death. This ADC-like approach leverages the stability of D-peptides to improve the therapeutic index of the payload. The drug is currently in preclinical development.
