Trimazosin is a small molecule drug classified as a selective alpha-1 adrenergic receptor antagonist (alpha blocker) and belongs to the n-arylpiperazine chemical class. It was originally developed by Pfizer and later licensed to Bristol-Myers Squibb for markets outside Canada, Mexico, and the USA. Trimazosin is primarily investigated for its antihypertensive effects in patients with mild to moderate essential hypertension and has also been considered for benign prostatic hyperplasia (BPH). Its mechanism of action involves blocking alpha-1 adrenergic receptors on vascular smooth muscle, leading to vasodilation and reduced systemic vascular resistance without significant sodium retention or renin axis stimulation. This pharmacological profile may contribute to sustained improvement in left ventricular function in patients with chronic congestive heart failure. Clinical studies have shown that it effectively lowers blood pressure with a favorable safety profile compared to other antihypertensives[1][2][3][4][5][7].
