Overview

GIC-1001 (trimebutine 3-thiocarbamoylbenzenesulfonate) is an orally administered, peripherally acting opioid agonist and hydrogen sulfide (H2S) donor developed by gIcare Pharma. It is designed to provide colonic analgesia and reduce spasms during procedures such as colonoscopy, potentially allowing for sedation-free examinations. The molecule is a novel salt of trimebutine, a known modulator of gastrointestinal motility and visceral sensitivity, combined with a 3-thiocarbamoylbenzenesulfonate moiety that releases H2S to enhance antinociceptive effects. Clinical development has primarily targeted visceral pain management in colonoscopy and irritable bowel syndrome (IBS).
