Trimer-4571 + 3M-052-AF + alum is an investigational combination vaccine regimen designed to prevent or treat HIV infection by eliciting broadly neutralizing antibodies (bnAbs) against the HIV envelope glycoprotein. The formulation consists of a stabilized, prefusion-clade A HIV envelope trimer protein (Trimer 4571, also known as BG505 DS-SOSIP.664), adjuvanted with both the synthetic TLR7/8 agonist 3M‑052‑AF and aluminum hydroxide (alum). Trimer‑4571 presents native-like conformations of the viral envelope to stimulate immune responses targeting conserved regions of the virus, while the dual adjuvant system enhances immunogenicity by promoting robust B-cell and T-cell activation as well as durable antibody production. The combination has demonstrated safety and immunogenicity in phase I clinical trials in healthy adults, inducing autologous tier‑2 neutralizing antibodies in some recipients[6][9]. It is being developed primarily for prophylactic vaccination but is also under investigation for therapeutic use in people living with HIV on suppressive antiretroviral therapy[5][6].
