Overview

Trimer44NMe is a small molecule polyamine transport inhibitor (PTI) designed to block the cellular uptake of exogenous polyamines, including putrescine, spermidine, and spermine. It is primarily utilized in a therapeutic strategy known as polyamine blocking therapy (PBT), which combines a PTI with an inhibitor of polyamine biosynthesis, such as alpha-difluoromethylornithine (DFMO). Because cells often upregulate polyamine transport to compensate for the loss of de novo synthesis, Trimer44NMe is essential for achieving profound intracellular polyamine depletion. Preclinical research has demonstrated that Trimer44NMe can effectively reduce the viability of gemcitabine-resistant pancreatic ductal adenocarcinoma cells, inhibit vascular smooth muscle cell proliferation to prevent restenosis, and impair the virulence of the fungal pathogen Candida albicans by targeting the fungal-specific transporter Hol1. It serves as a critical tool compound for investigating the role of polyamine metabolism in cancer, vascular disease, and infectious diseases.
