Trimethobenzamide is a small molecule antiemetic drug used to treat and prevent nausea and vomiting, particularly after surgery or due to gastroenteritis. It acts primarily as an antagonist of the dopamine D2 receptor in the chemoreceptor trigger zone (CTZ) of the medulla oblongata, thereby suppressing signals that induce nausea and vomiting. While its precise mechanism is not fully understood, it is believed to decrease activity in brain regions responsible for emesis. Trimethobenzamide has been available in oral capsule and injectable forms; rectal suppositories were withdrawn from the U.S. market due to lack of proven efficacy[1][2][3][4][5].
