Overview

Trimethyl resveratrol (3,4',5-trimethoxystilbene) is a methylated derivative of the natural polyphenol resveratrol. By replacing the hydroxyl groups of resveratrol with methoxy groups, the molecule achieves significantly enhanced lipophilicity and metabolic stability, addressing the poor bioavailability and rapid clearance typically associated with the parent compound. Preclinical research, particularly in models of Alzheimer's disease, has demonstrated that trimethyl resveratrol exerts potent neuroprotective effects by maintaining mitochondrial integrity and acting as a strong inducer of autophagy. Additionally, it modulates insulin signaling pathways, as evidenced by its effect on glycogen synthase kinase 3 (GSK3) phosphorylation, which helps counteract neuronal insulin resistance and oxidative stress. Its improved pharmacokinetic profile makes it a subject of significant interest for the treatment of neurodegenerative and metabolic disorders.
