TRIO-525 is a first-in-class bispecific antibody-drug conjugate (ADC), categorized as a Tumor Immunogenicity Enhancing Antibody Conjugate (TIE-ADC). It is designed to treat solid tumors, particularly triple-negative breast cancer (TNBC), by simultaneously targeting TROP2 on malignant cells and CD33 on myeloid-derived suppressor cells (MDSCs) within the tumor microenvironment. Mechanistically, TRIO-525 utilizes a unique antibody format engineered for high affinity to TROP2 and tumor-selective engagement of CD33, enabling the selective depletion of immunosuppressive MDSCs while sparing healthy hematopoietic and immune cells. By eliminating both cancer cells and MDSCs, TRIO-525 aims to reverse TME-driven immunosuppression and restore T-cell proliferation. Preclinical studies have demonstrated nanomolar potency, exceptional plasma stability, and dose-dependent tumor regression in xenograft models without significant off-target toxicity.
