Overview

Trioxsalen is a small molecule drug belonging to the psoralen class. It is a furanocoumarin derivative obtained from several plants, mainly *Psoralea corylifolia*. Trioxsalen acts as a photosensitizer and is used in combination with ultraviolet A (UVA) light in phototherapy (PUVA therapy) for the treatment of vitiligo—a condition characterized by loss of skin pigmentation—and sometimes hand eczema. The drug itself is pharmacologically inactive until it is photoactivated by UVA exposure; upon activation, it forms interstrand cross-links in DNA that can lead to programmed cell death (apoptosis) unless repaired by cellular mechanisms. This mechanism underlies its therapeutic effect but also contributes to risks such as increased skin cancer and premature aging of the skin. Trioxsalen was discontinued by manufacturers in December 2002 but remains notable for its historical use and ongoing research applications[1][3][5].
