Overview

Tripchlorolide is a natural small molecule compound derived from the plant Tripterygium wilfordii, structurally related to triptolide and known as Triptolide 12,13-chlorohydrin.[2][5][8] It exhibits potent **antitumor**, **anti-inflammatory**, and **neuroprotective** properties. Mechanistically, tripchlorolide induces cell death in various cancer cell lines, particularly A549 lung cancer cells, primarily by activating autophagy through inhibition of the PI3K/AKT/mTOR signaling pathway rather than triggering apoptosis.[1][4][5] Preclinical evidence also suggests it modulates inflammatory and immune pathways, exerts neuroprotective effects (including protection against dopaminergic degeneration in Parkinson’s and cognitive deficits in Alzheimer’s models), and inhibits microglial activation via the STING-NLRP3 pathway.[5][7][10]
