Triplatin is a polynuclear platinum compound (PPC) that represents a distinct class of platinum-based chemotherapeutics designed to overcome the limitations of mononuclear agents like cisplatin and oxaliplatin. It is characterized by a multi-platinum structure linked by flexible diamine chains, which enables it to form unique, long-range DNA adducts and exhibit a high affinity for polyanionic molecules such as glycosaminoglycans (GAGs). In pancreatic ductal adenocarcinoma (PDAC), triplatin exploits the GAG-rich, fibroblast-dense stroma to selectively accumulate within the tumor microenvironment. This localization enhances DNA damage and activates robust signaling pathways, including ATM/ATR and p53-mediated stress responses, effectively bypassing common mechanisms of platinum resistance. By remodeling the tumor-fibroblast niche and disrupting intercellular communication, triplatin offers a spatially targeted approach to treating desmoplastic cancers.
