Triplatin NC is a polynuclear platinum analog developed to address the pharmacokinetic limitations and toxicity associated with earlier platinum-based chemotherapeutics, such as cisplatin and the first-generation trinuclear platinum complex Triplatin (BBR3464). Unlike traditional platinum agents that form covalent adducts with DNA, Triplatin NC interacts with the DNA phosphate backbone through a non-covalent 'phosphate clamp' mechanism, leading to DNA condensation and inhibition of cellular processes. Preclinical studies conducted at Virginia Commonwealth University have demonstrated that Triplatin NC is effective in suppressing primary tumor growth and lung metastasis in breast cancer models, as well as peritoneal carcinomatosis in pancreatic cancer models, while also exhibiting anti-angiogenic properties.
