Triple-Mix is an investigational vaccine candidate developed by Bavarian Nordic in collaboration with the Battelle Memorial Institute, designed to provide protection against three major alphaviruses: Venezuelan equine encephalitis virus (VEEV), Western equine encephalitis virus (WEEV), and Eastern equine encephalitis virus (EEEV). The approach involves a mixture of three separate monovalent vaccines, each utilizing the non-replicating Modified Vaccinia Ankara - Bavarian Nordic (MVA-BN) viral vector platform. Each vector is engineered to express the structural envelope polyprotein (E3-E2-6K-E1) of its respective target virus. Preclinical studies in mice demonstrated that this 'Triple-Mix' strategy provides complete protection against lethal aerosol challenges from all three viruses. This research served as the foundation for the development of MVA-WEV, a single trivalent vaccine vector that incorporates antigens from all three viruses into a single MVA-BN backbone.
