**Triplet** is a preclinical, intratumorally administered lipid nanoparticle formulation containing three mRNAs encoding interleukin-21, interleukin-7, and membrane-bound 4-1BB ligand. Developed by etherna with academic collaborators, the mRNA-LNP therapy drives local production of these immunostimulatory proteins in the tumor microenvironment to enhance dendritic-cell activation, tumor-infiltrating CD8-positive T-cell responses, systemic antitumor immunity, and immunological memory. Preclinical studies reported activity in triple-negative breast cancer and immune checkpoint blockade-resistant cancer models.
