Tripod Therapeutics is developing an orally available small molecule drug substance designed to regulate the cellular localization of the proteasome for the treatment of cancer. This therapeutic approach, termed Proteasome Localization Control (PLC), focuses on modulating the spatial distribution of the proteasome within the cell rather than inhibiting its catalytic activity directly. By controlling where the proteasome is active—for instance, by preventing its translocation into the nucleus—the drug aims to disrupt the protein degradation pathways that cancer cells rely on for growth and survival. This mechanism is intended to provide a more targeted and potentially less toxic alternative to systemic proteasome inhibitors like bortezomib.
