Drug intelligence / Profile preview
TriPRIL CAR-T cells are autologous T lymphocytes genetically engineered to express a dual-targeted chimeric antigen receptor (CAR) incorporating a trimeric form of the natural ligand APRIL (a proliferation-inducing ligand). This trimeric ligand-based CAR is designed to co-target B-cell maturation antigen (BCMA) and transmembrane activator and CAML interactor (TACI) on malignant plasma cells. By targeting both BCMA and TACI, TriPRIL CAR-T cells aim to overcome antigen escape, a common resistance mechanism associated with monospecific BCMA-directed CAR-T therapies. The construct is fused to 4-1BB and CD3ζ intracellular signaling domains to promote T-cell activation and survival. Developed by researchers at Massachusetts General Hospital, TriPRIL CAR-T cells are currently being evaluated in a Phase 1 clinical trial for the treatment of relapsed or refractory multiple myeloma.
