Overview

Triprilumab (SKB336) is a novel trispecific monoclonal antibody developed by Kelun-Biotech. It is designed to simultaneously target three key pathways in the tumor microenvironment: programmed death-ligand 1 (PD-L1), cluster of differentiation 47 (CD47), and 4-1BB (CD137). By blocking the PD-1/PD-L1 and CD47/SIRPα "don't eat me" signals while providing agonistic stimulation to the 4-1BB costimulatory receptor on T cells and NK cells, triprilumab aims to enhance both innate and adaptive anti-tumor immune responses. This multi-pronged approach is intended to overcome resistance to traditional checkpoint inhibitors and improve efficacy in patients with advanced solid tumors.
