Tris + sodium chloride-Aeras-tuberculosis refers to a specific sterile buffer formulation consisting of 10 mmol/L tris(hydroxymethyl)aminomethane and 169 mmol/L sodium chloride. Developed by the Aeras Global TB Vaccine Foundation, this solution was utilized as the placebo control and antigen diluent in Phase I clinical trials for tuberculosis vaccine candidates, such as AERAS-404 (H4:IC31). Tris serves as a buffering agent to maintain a stable physiological pH, while sodium chloride ensures the solution is isotonic for injection. In clinical trial NCT01074801, it was administered via intramuscular injection to healthy, BCG-vaccinated, HIV-negative adults to evaluate the safety and immunogenicity of the active vaccine candidate against a baseline control.
