**Tris-CAR-T cells** are an autologous, locoregionally administered, tandem bispecific CAR-T-cell therapy for recurrent glioblastoma. Patient-derived T cells are lentivirally engineered to recognize the glioma stem cell-associated antigens **CD44** and **CD133**, with a truncated interleukin-7 receptor alpha intracellular-domain armor intended to support CAR-T-cell persistence and function. The product is being evaluated through intraventricular or intracavitary administration via an Ommaya reservoir in a Phase 1 study of recurrent glioblastoma.
