Overview

Trispecific chimeric antigen receptor (CAR) T-cell therapy is an engineered cellular immunotherapy designed to address tumor heterogeneity and improve T-cell persistence in solid tumors. In the specific context of the Tris-CAR-T program (NCT05577091), the therapy consists of autologous T-cells expressing a tandem CAR that targets two distinct glioma stem cell markers, **CD44** and **CD133** (Prominin-1), and is further "armored" with a modified **IL-7 receptor alpha (IL7Ra)** intracellular domain. This multi-targeting approach is intended to prevent antigen escape, while the IL7Ra signaling domain enhances the survival, expansion, and metabolic fitness of the CAR-T cells within the immunosuppressive microenvironment of recurrent glioblastoma (rGBM). Early clinical results from locoregional administration have demonstrated a favorable safety profile and evidence of radiographic tumor regression.
