TriSTAR (Tri-specific T cell Activation and Rejuvenation) is a platform of trispecific antibody-fusion molecules developed by Marengo Therapeutics for the treatment of refractory solid tumors. The TriSTAR architecture consists of a germline T-cell receptor (TCR) beta (Vβ) chain-targeting antibody fused to a costimulatory cytokine (typically IL-2) and an additional Fab arm directed against a tumor-associated antigen (TAA). This design allows for the selective activation and expansion of specific Vβ T-cell subsets, which are then retargeted to the tumor site. By combining TCR engagement, cytokine costimulation, and tumor targeting, TriSTAR molecules aim to promote potent and durable anti-tumor activity and induce immunological memory, potentially offering a superior alternative to conventional anti-CD3 T-cell engagers in immunologically "cold" tumors.
