TriSTAR0701 is a first-in-class trispecific T cell engager developed for oncology by Marengo Therapeutics. It employs a dual-payload antibody-fusion design that redirects specific Vβ T cell subsets to kill Nectin-4-expressing tumor cells. The molecule simultaneously provides targeted tumor engagement and synergistic T cell activation by targeting both the T cell receptor β chain and Nectin-4, aiming to overcome T cell exhaustion commonly seen in immunologically "cold" tumors. TriSTAR0701 is part of Marengo's TriSTAR platform, which features multi-specific antibody-fusion constructs derived from a proprietary library targeting germline-encoded variable regions of the TCR, fused to T cell co-stimulatory domains and tumor-associated antigen binders. The drug is currently in preclinical development and aims to deliver potent and durable anti-tumor immunity in solid tumors, especially those expressing Nectin-4[5].
