Overview

TriTEN antibody is a trispecific T-cell engager (TCE) currently in preclinical development by the Guangzhou Jinan Biomedical Research and Development Base for the treatment of cancer. The antibody is designed to simultaneously bind three targets: the CD3 epsilon subunit and the CD28 costimulatory receptor on T cells, and a tumor-associated antigen (TAA) on malignant cells. By engaging both CD3 (Signal 1) and CD28 (Signal 2), TriTEN aims to provide a more complete activation signal to T cells compared to traditional bispecific engagers, potentially enhancing anti-tumor efficacy, T-cell proliferation, and persistence within the tumor microenvironment. This trispecific approach is intended to drive potent, tumor-specific immune responses while minimizing systemic toxicity.
