Overview

TRIV-509 is a half-life extended monoclonal antibody developed by Triveni Bio that dually inhibits the serine proteases kallikrein 5 (KLK5) and kallikrein 7 (KLK7). These enzymes are overactive in atopic dermatitis (AD), leading to skin barrier dysfunction, inflammation, and pruritus. By inhibiting KLK5 and KLK7, TRIV-509 addresses three key pathological pillars of AD—restoring skin barrier integrity, reducing chronic inflammation, and alleviating itch. Preclinical models have shown superior efficacy of TRIV-509 compared to IL-4 receptor inhibition. It is currently in clinical development for moderate-to-severe atopic dermatitis[1][2][4][5][6].
