TRIV-573 is a half-life extended, second-generation bispecific antibody developed by Triveni Bio. It is designed to simultaneously inhibit kallikrein-related peptidase 5 (KLK5), kallikrein-related peptidase 7 (KLK7), and interleukin-13 (IL-13). KLK5 and KLK7 are serine proteases implicated in skin barrier dysfunction and inflammation, particularly in diseases like atopic dermatitis. IL-13 is a cytokine involved in allergic inflammation and fibrosis. By targeting both the proteases responsible for barrier breakdown and the cytokine driving Th2-mediated inflammation, TRIV-573 aims to provide comprehensive therapeutic benefits for immunological disorders characterized by barrier dysfunction and chronic inflammation. The drug is currently in preclinical/IND-enabling studies with initial focus on hereditary pancreatitis; it also has potential applications in atopic dermatitis and other inflammatory or immunological conditions[1][2][3][4][5][6][9].
