Overview

**TRK-001 + sirolimus** is a combination therapy composed of autologous expanded regulatory T cells (TRK-001) and the immunosuppressant sirolimus. TRK-001 is a cell therapy using the patient's own regulatory T cells (Tregs) to induce immune tolerance and reduce the need for traditional immunosuppressive medications post-organ transplantation. Sirolimus is a small molecule inhibitor of the mammalian target of rapamycin (mTOR), primarily used to prevent organ rejection by suppressing T cell activation and proliferation. This combination is being investigated in clinical trials for its ability to prevent kidney transplant rejection while potentially reducing long-term immunosuppression requirements. The therapy is designed so that patients receive TRK-001 and, after initial standard immunosuppression, transition to sirolimus monotherapy, aiming for reduced toxicity and improved graft survival[2][6][7].
