TRK-950 is a first-in-class humanized IgG1 monoclonal antibody that targets cell cycle associated protein 1 (CAPRIN-1), a novel cancer therapeutic target discovered by Toray. CAPRIN-1 is strongly expressed on the cell membrane of many solid tumors—including gastric, gastroesophageal junction (GEJ), bladder, and colorectal cancers—as well as on metastatic cancer cells and cancer stem cells, but is minimally expressed in normal tissues. By binding to CAPRIN-1, TRK-950 induces antitumor effects primarily through immune-mediated mechanisms such as antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies have demonstrated its potency in tumor models and safety in non-human primates. Clinical trials have shown promising efficacy—especially when combined with standard therapies like ramucirumab and paclitaxel—in patients with advanced or metastatic gastric/GEJ adenocarcinoma who express high levels of CAPRIN-1[2][3][6][8].
