Overview

This is a combination regimen consisting of four agents: - **TRK-950** is a humanized monoclonal IgG1 antibody targeting CAPRIN-1 (cytoplasmic activation/proliferation-associated protein-1), which is strongly expressed on solid tumors but not on normal tissues. It induces tumor cell death via fragment crystallizable-mediated effector functions, including antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cell-mediated phagocytosis (ADCP)[1]. - **Carboplatin** is a platinum-based chemotherapy that forms DNA crosslinks, disrupting DNA synthesis and transcription, leading to apoptosis in cancer cells. - **Pegylated liposomal doxorubicin** (brand name Caelyx) is a liposome-encapsulated formulation of doxorubicin, an anthracycline that inhibits topoisomerase 2, blocking DNA replication and cell division. The pegylated liposome prolongs circulation and enables enhanced delivery and retention within tumors[2][4]. - **Bevacizumab** is a monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and thus starving tumors of blood supply. This regimen leverages direct cytotoxicity (carboplatin, doxorubicin), targeted immune-stimulation and tumor cell killing (TRK-950), and anti-angiogenic effects (bevacizumab). TRK-950 has shown good safety, linear pharmacokinetics, and is under development for CAPRIN-1 expressing solid tumors; its combination with cytotoxic/immune-modulating drugs may be synergistic[1].
