This combination therapy consists of four agents: - **TRK-950:** A first-in-class humanized IgG1 monoclonal antibody targeting CAPRIN-1 (cytoplasmic activation/proliferation-associated protein-1), highly expressed on the surface of most solid tumor cells but not normal tissues. TRK-950 mediates tumor cell death mainly via antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cell-mediated phagocytosis (ADCP), leading to antitumor immune activation and macrophage infiltration. - **Gemcitabine:** A nucleoside analog small molecule chemotherapy agent that inhibits DNA synthesis, leading to cell death (antimetabolite). - **Carboplatin:** A small molecule, platinum-based chemotherapy agent that forms DNA crosslinks, inhibiting DNA replication and transcription, leading to apoptosis. - **Bevacizumab:** A humanized monoclonal antibody against VEGF-A (vascular endothelial growth factor A), blocks angiogenesis, and is used as an antineoplastic agent. This combination is under investigation for treatment of CAPRIN-1-expressing solid tumors, leveraging the immune-modulating and cytotoxic effects of the agents. Preclinical studies suggest synergistic antitumor efficacy for this combination and early-phase trials report safety and potential efficacy.
