TRK-950 + pegylated liposomal doxorubicin is an investigational combination therapy for advanced and metastatic solid tumors. TRK-950 is a first-in-class humanized monoclonal IgG1 antibody that targets the cancer-associated protein CAPRIN-1, which is strongly expressed on the cell membrane of many solid tumors. TRK-950 mediates tumor cell death via fragment crystallizable (Fc)-mediated immune functions, primarily antibody-dependent cell-mediated cytotoxicity and phagocytosis. It is being developed for use both as monotherapy and in combination with established chemotherapies. Pegylated liposomal doxorubicin is a reformulated version of doxorubicin, an anthracycline cytotoxic agent, encapsulated in polyethylene glycol-coated liposomes to enhance drug delivery and reduce toxicity. The combination aims to enhance anti-tumor immune and cytotoxic effects by leveraging both targeted immunotherapy and chemotherapy. Clinical investigation is ongoing, focusing on treatment-refractory metastatic or locally advanced solid tumors.
