TRK-950 + pembrolizumab is an investigational combination therapy for advanced solid tumors. TRK-950 is a first-in-class humanized IgG1 monoclonal antibody targeting cytoplasmic activation/proliferation-associated protein-1 (CAPRIN-1), which is highly expressed on most solid tumor cell membranes but not on normal tissue. TRK-950’s mechanism includes antibody-dependent cell-mediated cytotoxicity and phagocytosis, increasing tumor-infiltrating macrophages and inducing tumor cell death. Pembrolizumab is a programmed cell death protein 1 (PD-1) inhibitor, an immune checkpoint inhibitor that potentiates T-cell mediated antitumor responses. The combination is under investigation in Phase 1b clinical trials to assess safety, optimal dosing, and preliminary antitumor activity in various advanced solid malignancies, including after progression on prior therapies[1][2][3].
