TrkB bispecific T-cell engager is a novel immunotherapeutic agent in preclinical development designed to treat solid malignancies. It is a bispecific antibody construct that simultaneously targets Tropomyosin receptor kinase B (TrkB), a receptor tyrosine kinase often overexpressed in various cancers, and the CD3 epsilon subunit on T cells. The molecule was engineered using a standard 'knob-into-hole' IgG-like format, with targeting arms derived from human phage display libraries and B cells from chickens immunized with the TrkB extracellular domain (ECD). By bridging TrkB-expressing tumor cells and T cells, the engager induces T-cell activation and directed cytotoxicity, bypassing MHC restriction. This therapeutic approach specifically aims to address solid tumors where TrkB signaling is implicated in poor prognosis, suppression of anoikis, and the promotion of epithelial-mesenchymal transition (EMT).
