Overview

TRL345 is a native human monoclonal antibody developed by Trellis Bioscience for the prevention and treatment of human cytomegalovirus (HCMV) infections. It specifically binds with high affinity (approximately 50 pM) to the highly conserved AD-2 Site I epitope on glycoprotein B (gB) of HCMV, which is critical for viral entry into host cells. This binding results in potent neutralization of diverse clinical isolates and blocks infection in various cell types relevant to both transplant recipients and maternal-fetal transmission scenarios. The antibody has demonstrated superior binding affinity compared to other antibodies such as TCN-202 and has shown efficacy in preclinical models at preventing HCMV infection of placental cells, suggesting potential use in preventing congenital CMV transmission. Its mechanism involves inhibition of virus internalization and direct neutralization by targeting gB[3][4][5][7].
