TRN-008

TRN-008 (also known as TERN-201) is a potent, selective, and irreversible small molecule inhibitor of semicarbazide-sensitive amine oxidase (SSAO), also known as vascular adhesion protein-1 (VAP-1). Developed by Terns Pharmaceuticals (originally in-licensed from Eli Lilly), the compound was primarily investigated for the treatment of non-alcoholic steatohepatitis (NASH). SSAO is an enzyme highly expressed in the vasculature of the liver and is involved in inflammatory cell recruitment and oxidative stress, both of which drive the progression of NASH and fibrosis. In Phase 1 clinical studies, TRN-008 demonstrated significant and sustained inhibition of plasma SSAO activity. However, Terns Pharmaceuticals announced in 2022 that it would pause the development of TERN-201 to focus on other pipeline assets, such as its BCR-ABL and GLP-1 programs.
