Overview

TRN-257 is a controlled-release, low-sodium formulation of sodium oxybate being developed by Tris Pharma for the treatment of narcolepsy-associated cataplexy, excessive daytime sleepiness, and idiopathic hypersomnia. The drug utilizes proprietary RaftWorks and LiquiXR delivery technologies to enable a once-nightly dosing regimen, which is a significant improvement over the twice-nightly dosing required by standard oxybate treatments. Furthermore, TRN-257 is engineered to have a substantially lower sodium content (80 mg for a 9 g dose) compared to legacy products, addressing cardiovascular concerns associated with high sodium intake. As a sodium salt of gamma-hydroxybutyrate (GHB), its therapeutic effects are primarily mediated through agonist activity at GABAB receptors, modulating noradrenergic, dopaminergic, and thalamocortical neurons to improve sleep architecture and daytime alertness. The FDA accepted the New Drug Application (NDA) for TRN-257 in December 2025.
