Overview

TRO-02 is a conditionally activated antibody-drug conjugate (ADC) targeting the epidermal growth factor receptor (EGFR), developed by TriOar Inc. It utilizes the proprietary TROCAD™ and TROSIG™ platforms to enhance tumor selectivity and minimize on-target/off-tumor toxicities, such as skin toxicity, which often limit EGFR-targeted therapies. The ADC consists of the monoclonal antibody panitumumab conjugated to a potent topoisomerase I inhibitor with a drug-to-antibody ratio (DAR) of 8. TRO-02 is designed with a dual-function masking mechanism: it remains inactive until proteolytic cleavage by tumor-associated proteases and utilizes Annexin A1 targeting to facilitate transcytosis across tumor vasculature. Preclinical studies demonstrate high stability and potent antitumor efficacy in xenograft models.
