TROCEPT-X is a novel tumor-selective oncolytic virus platform based on a genetically engineered adenovirus type 5 (Ad5). It is designed to selectively target and replicate in αvβ6 integrin-positive epithelial cancer cells following intravenous administration. The virus has been rationally engineered to remove all natural tissue tropisms, thereby minimizing infection of normal tissues and reducing off-target toxicity. Upon infecting tumor cells, the platform enables in situ production of potent therapeutic payloads; the lead candidate encodes a fully human immune checkpoint inhibitor antibody for local expression within tumors. This approach aims to maximize anti-tumor efficacy while limiting systemic exposure and associated toxicities. Developed by Accession Therapeutics, the first-in-human clinical trial (ATTEST) is enrolling patients with various solid tumors including non-small cell lung cancer, bladder cancer, pancreatic cancer, head and neck cancers, cholangiocarcinoma, and endometrial cancers[1][2][3][8][10].
