Overview

Trontinemab is an investigational monoclonal antibody engineered using Roche's proprietary Brainshuttle technology to enhance delivery across the blood-brain barrier for the treatment of Alzheimer's disease. It is a reformulation of gantenerumab, modified with a transferrin receptor (TfR1) shuttle module that enables efficient transport into the central nervous system. Trontinemab targets aggregated forms of amyloid beta, binding and facilitating removal of amyloid plaques in the brain—a pathological hallmark of Alzheimer's disease. Clinical studies have shown that trontinemab achieves rapid and deep dose-dependent reduction in brain amyloid plaques, as well as significant decreases in downstream biomarkers such as total tau, phosphorylated tau (pTau181 and pTau217), and neurogranin in cerebrospinal fluid and plasma. The drug has demonstrated a favorable safety profile with low incidence of amyloid-related imaging abnormalities (ARIA). Trontinemab is being developed by Roche/Genentech for early-stage Alzheimer’s disease[1][2][4][5][6][7][8][9].
