Drug intelligence / Profile preview
Trop2 HCAb (Trophoblast cell surface antigen 2 heavy chain-only antibody) refers to a class of fully human single-domain or heavy chain-only antibodies targeting TROP2, a cell-surface glycoprotein overexpressed in various epithelial cancers (such as breast, lung, and colorectal cancers). Unlike conventional antibodies that consist of two heavy and two light chains, HCAbs lack light chains, resulting in a smaller antigen-binding domain (VH or VHH) that can access novel, hard-to-reach epitopes. This format is highly versatile and can be utilized as naked antibodies (such as KisoJi's KJ-103), engineered into multi-specific formats, or used as targeting domains in chimeric antigen receptor (CAR) T-cell therapies. Platforms like Nona Biosciences' Harbour Mice® and KisoJi's KisoMouse™ are used to discover and develop these high-affinity, fully human TROP2-targeting HCAbs to overcome resistance and toxicity challenges associated with traditional TROP2-directed antibody-drug conjugates (ADCs).
