**TROP2-MMAE protodrug** is a preclinical, click-activated cancer therapeutic system developed by Shasqi for **TROP2-expressing solid tumors**. The system comprises the TROP2-binding Fab activator SQT02, which carries tetrazine groups, and the separately administered MMAE protodrug SQP22, in which monomethyl auristatin E is attenuated and linked to trans-cyclooctene. Following tumor localization of the TROP2-directed activator, a tetrazine-trans-cyclooctene click reaction releases active MMAE locally, enabling microtubule-directed cytotoxicity while aiming to reduce systemic exposure relative to conventional antibody-drug conjugates.
