TROP2 ProTriTAC is a protease-activated T-cell engager prodrug developed by Harpoon Therapeutics for the treatment of solid tumors. Built on the proprietary ProTriTAC (Protease-activated Tri-specific T cell Activating Construct) platform, the molecule is a single polypeptide chain containing three humanized antibody-derived binding domains: anti-TROP2 for tumor targeting, anti-CD3 for T-cell recruitment, and anti-albumin for half-life extension. The drug is designed as a conditionally active biologic; it remains systemically inactive due to the masking effect of the albumin-binding domain. Upon reaching the tumor microenvironment, tumor-associated proteases cleave a specific linker, releasing the albumin-binding domain and masking moiety to reveal the active T-cell engager, which then facilitates T-cell mediated lysis of TROP2-expressing cancer cells.
